Abstract

Ä¡ÅÂÇü¼ºÀ» ¾ïÁ¦ÇÏ´Â À¯»ê±Õ K1270À» ±èÄ¡·ÎºÎÅÍ ºÐ¸®ÇÑ ÈÄ ¹è¾çÀû, »ýÈ­ÇÐÀû Ư¡ ¹× 16S rDNA¿°±â¼­¿­ ºÐ¼®¿¡ ÀÇÇØ Pediococcus pentosaceus K1270À¸·Î µ¿Á¤ÇÏ¿´´Ù. K1270 ±ÕÁÖ´Â 5% ÀÚ´çÀÌ ÇÔÀ¯µÈ ¹èÁö¿¡¼­ Streptococcus mutans Ingbritt¿¡ ÀÇÇÑ ÀΰøÄ¡Å Çü¼ºÀ» ´ëÁ¶±º¿¡ ºñÇØ 94.6% ¾ïÁ¦ÇÏ¿´À¸¸ç, ºñ¼ö¿ë¼º ±Û·çĵÀÇ ÇÕ¼ºÀ» 89.6% ¾ïÁ¦ÇÏ¿´´Ù. S mutans IngbrittÀÇ Áõ½ÄÀº ´ëÁ¶±º¿¡ ºñÇØ 100¹è Á¤µµ ¾ïÁ¦ÇÏ¿´´Ù. K1270±ÕÁÖ´Â TMB¿Í peroxidase°¡ ÷°¡µÈ MRS ÇÑõ¹èÁö¿¡¼­ °ú»êÈ­¼ö¼Ò¸¦ »ý»êÇÏ¿´À¸¸ç, ÀΰøÄ¡Å Çü¼º¿¡ ´ëÇÑ K1270±ÕÁÖÀÇ ¾ïÁ¦ È¿°ú´Â catalase ÷°¡¿¡ ÀÇÇØ ÀϺΠ°¨¼Ò µÇ¾ú´Ù. K1270 ±ÕÁÖÀÇ ¹è¾ç »óû¾×À» 10% ÀÚ´çÀÌ ÇÔÀ¯µÈ 2¡¿M17 broth¿¡ µ¿·® °¡ÇÑ °æ¿ì ÀΰøÄ¡Å Çü¼º ¹× Streptococcus mutans IngbrittÀÇ Áõ½ÄÀÌ ¾ïÁ¦µÇ¾úÀ¸¸ç, ÀÌ ¾ïÁ¦È¿°ú´Â catalase÷°¡¿¡ ÀÇÇØ ÀϺΠ°¨¼ÒµÇ¾ú°í, ¿­Ã³¸® , ¶Ç´Â trypsinóġ¿¡ ÀÇÇØ ¿ÏÀüÈ÷ ¼Ò½ÇµÇ¾ú´Ù. µû¶ó¼­, º» ¿¬±¸¿¡¼­ ºÐ¸®, µ¿Á¤µÈ p. pentosaceus K1270Àº °ú»êÈ­ ¼ö¼Ò¿Í bacteriocin À¯»ç¹°ÁúÀ» ºÐºñÇÏ¿© S. mutans IngbrittÀÇ Áõ½ÄÀ» ¾ïÁ¦ÇÔÀ¸·Î½á ÀΰøÄ¡Å Çü¼ºÀ» ¾ïÁ¦ÇÏ´Â °ÍÀ¸·Î »ç·áµÈ´Ù.

Pediococcus pentosaceus K1270 was isolated from naturally fermented kimchi and identified based on the 16S rDNA sequence as well as cultural and biochemical characteristics. This strain strongly inhibited the formation of biofilm by Streptococcus mutans Ingbritt. K1270 also showed antibacterial activity against S. mutans Ingbritt. It was observed that K1270 strain produced hydrogen peroxide on MRS agar supplemented with 3, 3, 5, 5¡þtetramethylbenzidine (TMB) and peroxidase and the inhibitory effect of K1270 strain on the biofilm formation was reversed by the addition of catalase in part. Culture supernatant of K1270 inhibited the biofilm formation and the multiplication of S. mutans Ingbritt. This inhibitory effect of culture supernatant was decreased slightly by the addition of catalase and abolished by heat or trypsin treatment. Thus, this study suggests that P. pen¡þtosaceus K1270 inhibit the biofilm formation through the inhibition of the replication of S. mutans Ingbritt by producing hydrogen peroxide and bacteriocin.
